For chemical profiling, plants were taken from axenic culture and subjected to organic extraction. The extracts were analyzed using gas chromatography-mass spectrometry (GC-MS). A total of 25 terpenoids were detected (Fig. 1), and retention indices of these compounds were calculated (Table 1). All of them are sesquiterpenes, including 7 unidentified compounds. AA6F87ECFFA54B0D61393FEAFA64FACE.taxon	description	To identify candidate terpene-producing genes in R. lindenbergiana, its transcriptomes from the One thousand plant (OneKP) project (https: // sites. google. com / a / ualberta. ca / onekp /) were searched for TPS and MTPLS genes using a HMMER method (Finn et al., 2015). A single TPS homolog designated as RlTPS 1 was identified. RlTPS 1 appears to be partial sequence. The protein it encodes is most similar to MpDTPS 5 from M. polymorpha (Supplemental Figure S 1). Because MpDTPS 5 is a putative diterpene synthase (Bowman et al., 2017), we hypothesized that RlTPS 1 is also a diterpene synthase. In contrast, a total of 12 MTPSL genes were found. Five of them that appeared to be full-length genes were designated RlMTPSL 1 - 5. A cDNA for each of the five genes was cloned by reverse transcription PCR (RT-PCR) and verified by sequencing. RlMTPSL 1 - 5 encode proteins of 391 – 441 amino acids in length (Fig. S 1). Sequence similarities among the five RlMTPSLs range from 38 % (RlMTPSL 1 and RlMTPS 4) to 71 % (RlMTPSL 2 and SlMTPSL 3). Sequence alignment showed that they had DDXXD and NSE / DTE conserved motifs, which were reported to be involved in ionization of the substrate and catalysis (Chen et al., 2011). AA6F87ECFFA24B0A626F3EFAFD7AFA6E.taxon	description	Plants of R. lindenbergiana were harvested for RNA extraction using the QIAGEN RNA isolation Kit. RNA was then converted to single-strand cDNA by GE First-strand cDNA synthesis Kit. The RlMTPSL genes were amplified by RT-PCR using the primers listed in Supplemental Table S 1. The amplification products were cloned into the protein expression vector pEXP 5 - CT / TOPO (Invitrogen) and were confirmed by sequencing. Protein expression constructs were transferred into E. coli BL 21 - Codon Plus (DE 3) for protein expression and the bacteria were cultured in LB medium at 37 ◦ until OD 600 reached 0.4 – 0.6. The recombinant proteins were expressed for 16 h at 18 ◦ C by adding 0.4 mM isopropyl-b-D- 1 - thiogalactopyranoside (IPTG). The cells were collected by centrifugation at 4000 g for 10 min. After that, they were disrupted by a 8 × 10 s treatment with a sonicator (Misonix Microson ultrasonic cell disruptor, USA) in chilled extraction buffer [50 mM Tris ⋅ HCl, pH 7.5, 5 mM DTT, 0.5 mM phenylmethylsulfonyl fluoride (PMSF), 10 % (vol / vol) glycerol].
